FLModafinil (also known as lauflumid or CRL-40,940) is a bis-fluorinated structural analogue of modafinil. In the molecule both phenyl rings of the benzhydryl fragment are substituted with a fluorine atom at the para position, which distinguishes it from the parent modafinil. Characterisation and research applications
FLModafinil differs from modafinil by the presence of two fluorine atoms at the para positions of both aromatic rings. The introduction of fluorine — an element of high electronegativity and small atomic radius — is a common strategy in medicinal chemistry, used to modify the electronic properties, lipophilicity and metabolic stability of a molecule without significantly altering its geometry. For this reason fluorinated analogues such as FLModafinil are valuable tools in structure–activity relationship (SAR) studies within the eugeroic class of compounds. Like modafinil, the molecule possesses a chiral centre at the sulfur atom of the sulfinyl group.
In the scientific literature FLModafinil is described as an experimental modafinil analogue studied for its wakefulness-promoting properties.
